2.6. Analysis of the histology These slices were examined with a light-optical microscope (Microscope BX51TF, Olympus, Tokyo, Japan) and evaluated semiquantitatively. Magnification of 100 and 200 times was used regularly. The evaluation of the histological slices of lung, liver, spleen and kidney is based on an evaluation scale, which uses a score from 0 to 2 to assign the following statements about the degree of the variances of the examined characteristics: “without noticeable results” (0), “minimal change” (1), or “middle to high degree of change” (2). The features which are only grouped in two categories, are assigned the score of 0 or 1. They are either categorized using the assessment of the spleen’s white to red pulp [“yes” (0, classifiable), or “no” (1, not classifiable)], or by the assessment of the abundance of the spleen’s white pulp and the kidney’s glomerulas “normal filled” (0) or “sparsely filled” (1). The assigned number could then be used for statistical evaluation. |
4.4.3 Auswertung der histologischen Präparate
Die für die histologische Auswertung angefertigten und mit Hämatoxylin und Eosin gefärbten Präparate der Organe (Lunge, Leber, Milz, Niere) wurden lichtmikroskopisch (Mikroskop BX51TF, Olympus, Tokyo, Japan) untersucht und semiquantitativ ausgewertet. Dazu wurden je nach zu bewertendem Kriterium 100- oder 200-fache bzw. für Detailansichten bis zu 1.000-fache Vergrößerungen eingesetzt. Die auf den histologischen Schnitten basierende Beurteilung erfolgte [page 59] nach einem Bewertungsschema (Scoringsystem), bei dem je nach Ausprägung eines Merkmals ein Zahlenwert (Score) von 0 bis 2 bei folgenden Aussagen über den Grad der Veränderungen des zu untersuchenden Merkmals vergeben wurde: „ohne besonderen Befund“ (0), „geringgradige Veränderungen“ (1) oder „mittel- bis hochgradige Veränderungen“ (2). Bei Merkmalen, die nur in zwei Kategorien eingeteilt wurden, wurde der Score 0 und 1 vergeben, wobei sich dieser entweder, wie bei der Beurteilung der Abgrenzung der weißen zur roten Pulpa der Milz, auf die Aussage „ja“ (0, abgrenzbar) oder „nein“ (1, keine Abgrenzung) bezog oder bei der Bewertung der Fülle der weißen Pulpa der Milz bzw. der Glomeruli der Niere auf „normal gefüllt“ (0) und „wenig gefüllt“ (1). The same results are presented, although Mallig (2006) is not mentioned anywhere and although a different population of experimental mice must have been used: According to the source, n=9 for the CCI group and n=10 for the Fx-Shock group as well as the Poly group. This is consistent with the figures, in particular the approx. 70% result for the Poly group. In the paper, however, it is claimed that n=18 for the CCI group and the Fx-Shock group as well as n=15 for the Poly group (see abstract), which is not consistent with the approx. 70% result for the Poly group. |
